Further electron microscope characterization of spore appendages of Clostridium bifermentans.
Two distinct spore appendage types of Clostridium bifermentans, a pinlike appendage and a tubular appendage, were studied by electron microscopy. The pinlike appendage is characterized by a shaft, about 100 A in diameter, which has a lobed caplike structure. The tubular appendage, 500 to 600 A in diameter, is characterized by a hirsute region consisting of small filaments or fibrils. Gross morp...
متن کاملThe distribution of cholinesterase in cholinergic neurons demonstrated with the electron microscope.
The thiocholine technique for cholinesterase has been successfully adapted for the demonstration of enzyme distribution with the electron microscope. After fixation in buffered glutaraldehyde, thin slices of tissue were taken through a histochemical procedure designed to minimize diffusion artifacts and cytological damage; appropriate areas were dissected out, fixed with osmium tetroxide and em...
متن کاملthe scanning electron microscope
it is only thirteen years since the scanning electron microscope has been available commercially. yet, even in this short period of time, this instrument has been a powerful tool in the investigation of topography, electrical and magnetic properties, crystal structure, cathodoluminescent characteristics etc. of solid specimens. today, this type of microscope has opened its place alongside the c...
متن کاملAutoradiography with the Electron Microscope
Autoradiographic techniques for use with the electron microscope have been developed and considerably improved in recent years. A review of this historical development is given by Meek and Moses (14), and a theoretical analysis of the problems involved by Pelc (17) and Caro (4). In this paper, we describe a method for preparing autoradiographic specimens, using a new, fine-grained emulsion and ...
متن کاملIntracellular localization of nigeran in the wall of Aspergillus aculeatus by autoradiography with the electron microscope.
Aspergillus aculeatus mycelium was incubated with [(3)H]glucose under conditions that promote nigeran (mycodextran) accumulation (low pH, in the absence of nitrogen). Autoradiography revealed that essentially all the (3)H was localized around the hyphal perimeter. The results strongly support a hyphal wall location for nigeran.
